Les Motivations

 Les molécules biologiques ainsi que certains polymères bioinspirés sont des molécules flexibles, c'est-à-dire existant sous plusieurs conformations. Leur étude en phase gazeuse, en absence d'environnement, permet notamment :

ˇ          d'isoler les conformations les plus stables

ˇ          de les identifier en caractérisant les interactions qui les stabilisent, notamment par le réseau de liaisons hydrogène qui les structure

ˇ          puis de documenter la dépendance des propriétés de la molécule en fonction de sa conformation, notamment le lien entre structure et réactivité.

 Après des études structurales de molécules biologiques, comme des modèles de chaines peptidiques, qui nous ont permis d'isoler et de caractériser les structures secondaires typiques des protéines en phase gazeuse , les études actuelles visent à caractériser

ˇ          Sur des peptides modèles complexes , les interactions sous-tendant la structure locale des protéines, avec notamment un effort soutenu concernant i) l influence de l hydratation sur les propriétés structurales, notamment la flexibilité, ii) l influence d hétéroatomes, dont l'interaction avec l'environnement proche dans une protéine est assez peu documentée, ou encore iii) le rôle des résidus aromatiques contigus dans la formation spontanée de domaines hydrophobes.

ˇ          Sur des molécules biomimétiques, comme les foldamères, oligomères peptidiques synthétiques modifiés par le chimiste pour obtenir des propriétés de repliement spécifiques (formation d hélices ou plus original de rubans, susceptibles de mimer une portion de protéine), les interactions stabilisant les briques élémentaires de ces foldamères, en particulier celles s'appuyant sur la présence d un hétéroatome comme le soufre, et leurs combinaisons, dont la connaissance permet d'orienter la synthèse vers les polymères les plus intéressants pour les applications.

ˇ          Sur des petits peptides modèles, la relaxation électronique suite à l'excitation UV des acides aminés aromatiques présents dans les protéines: ici l'enjeu, dans le contexte de photodommages aux systèmes vivants, est la connaissance des mécanismes de relaxation, leur dépendance avec la conformation et la modélisation par chimie quantique des états excités.

Foldamères et liaisons-H : le projet TUNIFOLDS

Le but de ce projet est i) de concevoir de nouvelles briques moléculaires pour foldamères, dont la flexibilité peut etre finement ajustée grace à la présence d'une fonction thioéther aliphatique, puis ii) d explorer leur capacité à induire de nouvelles topologies de repliement, dans des structures qui d ordinaire ne se replient pas, ou à promouvoir des architectures de repliement alternatives dans de petits oligomères. L originalité de ce projet, qui réunit chimistes organiciens, spectroscopistes et chimistes théoriciens, repose dans la combinaison unique de i) l exploitation des propriétés spécifiques du soufre à induire des interactions non-covalents dans les oligomères, ii) la caractérisation des conformations de foldamères modèles grace à des techniques spectroscopiques de la phase gazeuse sélectives en conformation, encore peu utilisées dans le domaine des foldamères, ainsi qu au VCD en solution, et iii) du role de la théorie pour sélectionner les briques les plus appropriées.

Modèles de chaines peptidiques

La motivation des études sur les peptides et les polymères synthétiques repose sur la collecte de données spectroscopiques précises permettant la validation des nombreuses simulations existantes sur les petits peptides, molécules tests avant la simulation des protéines. Les petits peptides étudiés, protégés à leurs extrémités, sont des modèles de segment de protéine et permettent donc d aborder des thématiques essentielles telles que la compétition entre les différentes structures secondaires des protéines comme les feuillets b, les coudes b ou g ou encore les hélices a.

Plus récemment, les études récentes ou en cours, notamment spectroscopiques, se concentrent sur les divers types d'interations au sein des proteines et leur lien avec la stabilisation des structures secondaires, avec pour cibles: - les hydrates de résidus modèles comme la phénylalanine, - les résidus polaires présentant une grande richesse de modes d'interaction avec leur environnement (Histidine, Asparagine, Aspartate) - les résidus plus rares comme la méthionine, présentant des interactions plus exotiques dans les biomolécules comme les liaisons hydrogène ayant un atome de soufre pour accepteur, ou encore des paires ioniques modélisant un résidu chargé négativement (par ex. carboxylate) interagissant avec cation métallique. Parallèlement, est également ciblée la photophysique des résidus aromatiques au sein de chaines peptidiques modèles, comme la phénylalanine.

Létude de la solvatation moléculaire léchelle microscopique a été un sujet de recherches trés prisé au début des années 90 notamment avec des solvants modéles comme les gaz rares. Le systéme benzéne-argon par sa simplicité a été emblématique de toute une série d’études avec diverses molécules aromatiques et  divers solvants. Les publications associées ont fait références pour les études des agrégats de gaz rares dopés.
