Certains micro-organismes monocellulaires tels que les bactéries magnétotactiques synthétisent et organisent des structures nanocristallines. L'objectif du stage est de développer un système microfluidique permettant de capturer une seule bactérie afin de l’étudier.
Some single cell micro-organisms such as magnetotactic bacteria synthesize and organize nanocrystalline structures. We aim to develop a microfluidic system that allows to capture a single bacteria in order to study it.

Le contexte
Certains micro-organismes monocellulaires tels que les bactéries magnétotactiques synthétisent et organisent des structures nanocristallines, la chaîne du magnétosome, dans un environnement physiologique avec un degré de contrôle bien supérieur à celui obtenu par les chimistes dans des conditions difficiles (température / pression élevée et solvants organiques). Cette biominéralisation implique généralement le transport de quantités infimes de produits chimiques (femtogrammes) du pool de substances labiles, aux compartiments intracellulaires dans lesquels ils sont cristallisés et organisés.
La mise au point de méthodes d’encapsulation utilisant la microfluidique permet de piéger des cellules uniques et de surveiller de manière dynamique la quantité physiologiquement pertinente de produits chimiques (de l'ordre du femtogramme - fg) à des concentrations (micromolaire - µM) dépassant de loin la limite de détection des techniques analytiques conventionnelles. De plus, des environnements de taille similaire à celle de la bactérie peuvent être produits, ce qui permet de modifier la morphologie de la cellule et son organisation intracellulaire. Ces processus peuvent être analysés à haut débit et dans des études parallélisées.

Mission
Le candidat retenu contribuera à la conception, la microfabrication et l'optimisation du microsystème, ainsi qu'aux campagnes de mesure et à l'analyse des données. En particulier, il aura à déterminer l’influence du milieu chimique et de l’environnement physique sur la nucléation et l’organisation des magnétosomes au niveau d'une cellule unique. Des techniques de microfluidique et de microscopie quantitative corrélative seront utilisées à cet effet. Un accent particulier est mis sur la recherche interdisciplinaire dans le cadre d'une collaboration étroite avec les scientifiques travaillant sur les aspects chimiques, biologiques et physiques de la biominéralisation et de la biomimétique des assemblages magnétiques.

Le candidat travaillera dans l'équipe de recherche LIONS (CEA Saclay), en étroite collaboration avec les biologistes du BIAM (CEA Cadarache). Un séjour à Cadarache pour mieux connaitre la culture cellulaire est possible.

Profil
Nous recherchons un étudiant ayant une formation en chimie, biophysique, biotechnologie ou physique.
Des compétences en microfluidique, en chimie analytique et en microscopie à fluorescence sont appréciées. Des compétences en programmation et en conception CAO sont un avantage.
Une bonne connaissance de l'anglais est requise, ainsi qu'une forte motivation personnelle et de la fiabilité.
Context
Some single cell micro-organisms such as magnetotactic bacteria synthesize and organize nanocrystalline structures, the magnetosome chain, in physiological environment with a degree of control far superior to that obtained by chemists using harsh conditions (high temperature/pressure and organic solvents). This so-called biomineralization typically involves transport of minute amounts of chemicals (femtograms) from the labile pool to intracellular compartments in which they are crystallized and organized.
The development of encapsulation methods using microfluidics allows single cell trapping and dynamic monitoring of physiologically relevant amount of chemicals (fg) to be contained at concentrations (µM) typically far exceeding the detection limit of conventional analytical techniques. In addition, environments with a similar size to that of the bacteria can be produced such that the morphology of the cell and its intracellular organization can be altered. Such processes can be analyzed with high throughput and in parallelized studies.

Mission
The successful candidate will contribute to the design, the microfabrication, and the optimization of the microsystem, to the measurement campaigns and to the data analysis. Especially he/she will determine the influence of the chemical and the physical environment on the nucleation and organization of the magnetosomes at the single cell level. Microfluidics and correlative quantitative microscopy techniques will be used to this effect. A special emphasis is laid on interdisciplinary research so that close collaboration with scientists working on chemical, biological and physical aspects of biomineralization and biomimetics of magnetic assemblies will be expected.
The candidate will work at LIONS (CEA Saclay) but he/she will be in close collaboration with the biologists from the BIAM (CEA Cadarache). A stay in Cadarache to learn about cell culture is possible.

Profile
We are seeking a student with a background in chemistry, biophysics, biotechnology, or physics.
Skills in microfluidics, analytical chemistry, and fluorescence microscopy are appreciated. Programming skills and CAD design would be a plus.
Good knowledge in English, reliability and self-motivation are required.
