Étude du greffage covalent de colorants pour la conception d’une optode pour la mesure du pH

Résumé :

Cette étude s’inscrit dans le domaine des capteurs pour la mesure du pH, et plus particulièrement des capteurs chimiques à fibre optique, également appelés optodes. Bien que ce type de capteur optique existe déjà, les optodes disponibles peuvent être améliorées, notamment leur stabilité dans le temps et leur durée de vie. Pour ce faire, une piste intéressante consiste à remplacer l’immobilisation physique ou électrostatique du(des) indicateur(s) coloré(s) de pH sur la fibre optique classiquement utilisée par un greffage chimique covalent. Cette étude consiste ainsi à étudier le greffage covalent de colorants pour la conception d’un nouveau type d’optode.

Dans un premier temps, le procédé de greffage GraftFastTM a été sélectionné parmi les nombreuses techniques de greffage chimique décrites dans la littérature. Ce procédé, développé au sein de notre laboratoire, est basé sur la réduction par un agent chimique de sels d’aryldiazonium afin de former des radicaux aryles, pouvant se greffer sur tout type de substrat. Il permet d’obtenir rapidement des films polymériques sans préparation préalable du substrat. Ensuite, deux colorants - le rouge neutre et la rhodamine 560 - ont été sélectionnés et leurs propriétés spectroscopiques ont été étudiées.

Dans un second temps, ces deux colorants ont été greffés sur le substrat de référence pour le procédé GraftFastTM, à savoir des lames d’or. Le greffage de ces deux colorants a été caractérisé (par spectrophotométrie UV-Visible, par spectroscopies infrarouge et de photoélectrons induits par rayons X et par microscopie à force atomique) et optimisé par l’étude paramétrique des conditions expérimentales. Ensuite, une méthode expérimentale innovante, dite de séquençage, a été mise au point et testée afin d’augmenter l’efficacité du greffage.

Enfin, le greffage sur deux nouveaux substrats - des lames de PMMA et des lames d’or semi-transparentes - a été étudié, afin de transposer les connaissances fondamentales sur le greffage des colorants par le procédé GraftFastTM acquises précédemment au cas de capteurs réels à base de fibres optiques.

Study of the covalent grafting of dyes for the design of an optode for pH measurement

Abstract:

This study concerns sensors for pH measurement, and more precisely fiber optic chemical sensors, also called optodes. Although this kind of optical sensor already exists, the performance of the available optodes can be improved, especially their stability over time and their lifetime. To this end, an interesting idea is to replace the conventional immobilization by physical entrapment or electrostatic binding of the pH indicator(s) on the optical fiber by a chemical covalent grafting. The purpose of this work is to study the covalent grafting of dyes for the design of a new type of optode.

Firstly, the grafting process GraftFastTM was selected amongst the many chemical grafting techniques described in the literature. This process – developed in our laboratory – is based on the reduction of aryldiazonium salts by a chemical reagent, so as to create aryl radicals which can be grafted on any substrate. Polymeric films can be quickly obtained with this process without specific substrate preparation. Then, two dyes – neutral red and rhodamine 560 – were selected, and their spectroscopic properties were studied.

Secondly, the dyes were grafted on the preferred substrate for the GraftFastTM process, namely gold lamellas. The grafting of those dyes was characterized – by UV-Visible spectrophotometry, infrared and X-ray photoelectron spectroscopies and atomic force microscopy – optimized by the parametric study of the experimental conditions. Then, an innovative experimental method, called sequencing, was developed and tested in order to increase the efficiency of the grafting.

Finally, the grafting on two new substrates – PMMA lamellas and see-through gold lamellas – was studied, in order to implement the fundamental knowledge about the grafting of the two dyes by the GraftFastTM process previously acquired in the case real sensors based on optical fibers.
